ABSTRACT

Oligonucleotide drugs have the characteristics of targeting, modifiability and high biosafety. Recent studies have shown that oligonucleotide can be used to make biosensors, vaccine adjuvants, and has the functions of inhibiting alveolar bone resorption, promoting jaw and alveolar bone regeneration, anti-tumor, destroying plaque biofilm, and precise control of drug release. Therefore, it has a broad application prospect in the field of stomatology. This article reviews the classification, action mechanism and research status of oligonucleotide in stomatology. The aim is to provide ideas for further research and application of oligonucleotide.

ABSTRACT

The malignant tumors including oral cancer, colorectal cancer, pancreatic cancer, and esophageal cancer, of the digestive system are a common high-fatal malignancy. Porphyromonas gingivalis, as the most important pathogen of periodontal disease, has been gradually proved that its invasiveness occurs not only in the mouth but also in other parts of the digestive system. Moreover, the relevant pathogenic mechanism is increasingly attracting the reseachers' attention. In this study, the role and possible pathogenesis of Porphyromonas gingivalis in the digestive system are described in a systematic and comprehensive way.

ABSTRACT

Objective To in silico clone the NAD1 gene of three common parasites and analyze their bioinformatics,so as to lay the foundation for further research on the NAD gene. Methods By using the in silico cloning method,the full length cDNA (s)of NAD 1 genes of Clonorchis sinensis,Ascaris lumbricoides and Schistosoma japonicum were got,then their physical and chemical properties,compositions of amino acids,subcellular localizations,binary and ternary structures were contrastively an-alyzed.Results The three kinds of NAD1 proteins were similar in the relative molecular weight,subcellular localization,and physical and chemical properties.The NAD1 proteins were highly similar in binary and ternary structures of A.lumbricoides and S.japonicum.The phylogenetic analysis showed that C.sinensis,A.lumbricoides and S.japonicum belonged to the different evolu-tionary branches with a certain of genetic distance. Conclusion The three NAD1 genes got from C.sinensis,A.lumbricoides and S.japonicum by in silico cloning belong to the same gene of different species,which can be widely used in the researches of heritable variation of parasites.

ABSTRACT

Flavonoids have attracted much attention due to their good anti-inflammatory, anti-oxidation and anti-tumor effects. At present, the extraction of flavonoids is mainly based on organic solvent, while the researches on the use of green and safe solvents are quite limited. Therefore, in the present study, different types of deep eutectic solvents (DESs) were applied to investigate their effect on extraction of flavonoids and optimize the process, also investigate the recovery efficiency of DESs and evaluate the recovery method for total flavonoids. The extraction yield of the total flavonoids acted as the comprehensive evaluation indexes, and a central composite design (CCD) of response surface methodology (RSM) was employed to further optimize the alcohol-based DES extraction conditions. The results showed that the optimized extraction conditions were as follows: water-DES ratio of 27%, solid-liquid ratio of 15 mL·g⁻¹, extraction temperature of 83 °C and extraction time of 42 min in ChCl-glycerol at 1:4 ratio. Under these conditions, the mean experimental value of the extraction yield (75.05 mg·g⁻¹) corresponded well with the predicted value (77.86 mg·g⁻¹). Moreover, these experimental results showed more advantages such as in higher efficiency, economy and environmental protection as compared with previously reported conventional extraction methods. In addition,the recovery yield of the total flavonoids from the DESs extraction solution achieved 97.88% by using AB-8 macroporous resin, and 88.12% desorption ratio can be achieved by 100% ethanol with 5 times resin content. After the above treated DESs were collected, the extraction yield with the same method reached 95.23%, indicating that the method of macroporous resin can be used for efficient and simple recovery and reuse. This study suggests that DESs can be used as a kind of sustainable and efficient natural extraction solvents for extraction of flavonoids from Prunella vulgaris.

ABSTRACT

Prunellae Spica is a perennial edible and medicinal plant, rich in antioxidant substances. Total flavonoids (TFC), Phenolics (TPC), triterpenoids (TSC), polysaccharides (PC) and their antioxidant capacities (by the FRAP, DPPH and ABTS⁺ methods) of ethyl acetate fraction, n-butanol fraction and other fractions of aqueous extract from Prunellae Spica were investigated in this study. Then the multivariate statistical method was adopted to analyze the relationship between the multiple pharmaceutical ingredients and antioxidant capacities of Prunellae Spica. The results showed that ethyl acetate fraction had relatively high concentration of TFC (0.61±0.10) g·g⁻¹DW, TPC (0.52±0.09) g·g⁻¹DW, and TSC (0.21±0.03) g·g⁻¹DW, with high scavenging capacity of DPPH (3.1±0.38) mmol·L⁻¹·g⁻¹DW and FRAP (2.56±0.35) mmol·L⁻¹·g⁻¹DW. Hierarchical clustering analysis (HCA) and principal component analysis (PCA) results indicated the information from chemical compositions and antioxidant capacity can represent the "differences" of different fractions. Canonical correlation analysis (CCorA) revealed a high positive correlation between the amounts of multiple chemical compositions and the antioxidant capacities (r=0.970 0), and the first canonical variate had been reached. Moreover, ABTS⁺ method showed a low response to the compositions of different fractions, so this method may not be suitable for evaluation of Prunellae Spica antioxidant capacities, while DPPH evaluation method was more suitable for TSC and TPC. The results of this study have important reference significance for the evaluation method on antioxidant activity of Prunellae Spica in the field of food or medicine as well as for the development of related extracts.

ABSTRACT

OBJECTIVE: To evaluate the effect of supernatants of alcohol extracting-water precipitation of Polygonatum odoratum(POS) on hyperthyroidism-yin deficiency rats. METHODS: SD rats were randomly divided into control, model and POS groups(400 mg·kg-1). The hyperthyroidism-yin deficiency rats were treated by intraperitoneal injection of T3(500 μg·kg-1) for a week. The content of T3, T4 in serum and cAMP, cGMP in plasma was determined. A GC-MS based metabolic profiling method was employed to analyze the distinctive metabolic patterns of serum in control, model and POS. Multivariate statistical analysis was applied for multivariate analysis to find potential biomarkers of hyperthyroidism-yin deficiency by SIMCA-P 12.0 software. RESULTS: Compared with the model group, POS group can significantly reduce the content of T3, T4, cAMP, the ratio of cAMP/cGMP and increase the content of cGMP, indicating it has certain Yin-nourishing action. The PCA and PLS-DA score plots showed metabolic profiling of control, model and POS had significant differences, but which could be regulated nearly to normal state after POS interference. Four potential biomarkers were identified by PLS-DA and variable importance for project values(VIP) based on the metabolomics. They were 3-hydroxybutyric acid, galactose, palmitic acid and cholesterol. CONCLUSION: POS may play a role in treating hyperthyroidism-yin deficiency rats, by adjusting energy metabolism, glucose metabolism and lipid metabolism. The changes of metabolic pathways revealed by this study may provide some basis for the diagnosis and treatment of hyperthyroidism Yin-deficiency.

ABSTRACT

OBJECTIVE: To investigate the changes of platelet mitochondrial adenosine triphosphate synthase 6( ATPase-6)gene in patients with occupational chronic myeloid leukemia( CML) caused by benzene. METHODS: Five occupational benzene induced CML patients were selected as the case group and 21 healthy workers without benzene exposure were selected as the control group,venous blood were drew from these study subjects. The number of platelet was counted. The level of mitochondrial ATPase-6 gene was examined by quantitative polymerase chain reaction. RESULTS: The platelet count in case group was lower than that of control group [( 135. 2 ± 32. 6) ×10~9/ L vs( 239. 7 ± 77. 5) ×10~9/ L,P < 0. 05]. The ratio of ATPase-6 gene / platelet was higher than that of control group( median: 3. 43 ×10~(-4)copies vs 1. 87 ×10~(-4)copies,P < 0. 05). The ATPase-6 gene level in case group showed no significant difference when compared with that of control group( median: 4. 83 ×10~7 copies / L vs 3. 24 ×10~7 copies / L,P > 0. 05). CONCLUSION: The benzene induced CML patients shows more sensitive change in ratio of ATPase-6 gene / platelet than in ATPase-6 gene level.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of crosslinks on the stability of the spine and pedicle screws.</p><p><b>METHODS</b>Compression fracture of the L1 vertebra was produced in 30 fresh thoracic and lumbar vertebrae samples obtained from adult sheep, which were divided into 3 groups (n=10)with lot-drawing method. Four screws were fixed onto the superior and inferior pedicles of vertebral arch close to the fractured vertebrae, with different number of crosslinks (0 in Group A,1 in Group B, and 2 in Group C) on the rods. After fixation, the samples were subject to 10 000 times of fatigue test with 1.5 Hz load on the HY-3080 computer-control electronic universal test machine and HY-1000NM computer-control torsion test machine. The axial compressive stiffness, maximum pullout strength,and range of motion (ROM) of 6 directions, i.e., flexion, extension, left and right lateral bending, and left and right axial rotation of the 3 groups were measured and compared.</p><p><b>RESULTS</b>There were no statistically significant differences in axial compressive stiffness as well as the ROM of flexion, extension, and left and right lateral bending (all P>0.05). The maximum pullout strength was significantly smaller in Group A and Group B than in Group C [(129.56±29.63)N vs.(294.67±23.25) N,P=0.000;(254.02±36.29)vs.(294.67±23.25)N, P=0.006]. The ROM of left axial rotation was the highest in Group A(13.35°±1.06°), followed by Group B(12.23°±1.06°)and Group C (11.04°±0.74°)(F=13.44, P=0.000; Group B vs. Group A, P=0.000; Group B vs. Group C, P=0.001; Group C vs. Group A,P=0.000). The ROM of right axial rotation was also the highest in Group A(13.56°±1.15°), lower in Group B (12.39°±1.01°) and the lowest in Group C (10.81°±0.51°) (F=21.91, P=0.000; Group B vs. Group A,P=0.002; Group B vs. Group C, P=0.001; Group C vs. Group A, P=0.000).</p><p><b>CONCLUSION</b>Crosslinks may reinforce the pullout strength of the screws and improve the axial stability of the spine.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the clinical effects of cervical posterior pedicle screw internal fixation and interbody fusion for the treatment of atlanto-axial vertebral instability fractures.</p><p><b>METHODS</b>From July 2008 to July 2013, 21 patients with atlanto-axial vertebral instability fractures were treated with vertebral pedicle screw internal fixation and interbody fusion through posterior approach. There were 14 males and 7 females, aged from 20 to 55 years old with an average of 32 years. Lifted and guided the atlanto-axial vertebral pedicle screw to reduce atlanto-axial vertebral displacement.</p><p><b>RESULTS</b>All patients were followed up from 6 to 24 months with an average of 12.5 months. Wounds got healed without complication of infection. Clinical symptoms were relieved at 6 months after operation. According to ASIA score standard to assess at 6 months after operation, the items of motion, light touch and needle score had obviously improved, and respectively were 99.45 ±0.27, 111.09 ± 0.47,111.11 ± 0.58. VAS and NDI scores also had obviously improved, and respectively were 1.04 ± 0.38 and 12.56 ± 2.24. Imaging examinations showed internal fixation locations were good, without atlanto-axial joint instability.</p><p><b>CONCLUSION</b>Cervical posterior pedicle screw internal fixation and interbody fusion can effectively restore the stability of atlanto-axial joint, reduce complication and obtain satisfactory effects.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the impact of pedicle cortex perforation on the stability of internal fixation of the vertebral body fracture, and to compare the stability of the vertebrae with pedicle cortex perforation after the injured vertebra transpedicular screw fixation by different ways.</p><p><b>METHODS</b>A total of 36 fresh thoracic and lumbarvertebrae samples of adult sheep (T₁₃-L₁) were equally divided into four groups (A, B, C and D) by using the random number table method. The vertebral compression fracture was performed in the L1 vertebral body of the four groups using the Chiba's method. Four pedicle screws were fixed on the upper and lower injured vertebrae of four groups. In addition, the group C was fixed into a pedicle screw through the injured vertebrae; D group was set two pedicle screws through the injured vertebrae. Then the samples of group B, C, and D were removed a quarter of either side of lateral T₁₄ thoracic pedicle, which was considered as the pedicle cortex perforation model. Four groups were performed fatigue test of 10 000 times by (300 ± 105)N load. The drawing force of the screw and the stability of injured thoracolumbar vertebrae were measured and the differences in every group were compared.</p><p><b>RESULTS</b>The axial compressive stiffness and maximum drawing force of screws in the other three groups were significantly higher than those in group B (all P=0.000). The maximum range of motion in four directions of group B were significantly larger than those of the other three groups (all P=0.000). The stiffness and the drawing force in groups C and D were significantly larger than those in the group A (all P=0.000), and the maximum range of motion in four directions of the two groups were lower than that of group A (P=0.002, P=0.005). Every testing indicator in group C had no significant difference when compared with group D (P>0.05).</p><p><b>CONCLUSIONS</b>The pedicle cortex perforation seriously affects the stability of the fractured vertebral body. The injured vertebra transpedicular screw can increase the stability of internal fixation.</p>

ABSTRACT

<p><b>BACKGROUND</b>It has been suggested that glycated hemoglobin (HbA1c) underestimate the actual glycemic control levels in maintenance hemodialysis (MHD) patients, because of anemia and the using of erythropoietin (EPO); it was recommended that glycated albumin (GA) should be an alternative marker. Therefore, the assessment performances of glycemic control were compared between GA and HbA1c in this research by referring to mean plasma glucose (MPG) in diabetes mellitus (DM) patients undergoing MHD or not.</p><p><b>METHODS</b>MPG was calculated according to the data registered at enrollment and follow-up 2 months later and corresponding HbA1c, albumin (ALB), GA, etc. were measured in 280 cases. A case-control study for comparing GA and HbA1c was done among the groups of MHD patients with DM (n=88) and without DM (NDM; n=90), and non-MHD ones with DM (n=102) using MPG for an actual glycemic control standard.</p><p><b>RESULTS</b>In these 3 groups, only for DM patients' (whether undergoing MHD or not), GA and HbA1c correlated with MPG significantly (P < 0.01). Through linear regression analysis, it could be found that the regression curves of GA almost coincided in MHD and non-MHD patients with DM, because the intercepts (2.418 vs. 2.329) and slopes (0.053 vs. 0.057) were very close to each other. On the contrary, regression curves of HbA1c did not coincide in the two groups, because variance of the slopes (0.036 vs. 0.052) were relatively large. Through comparing receiver operating characteristic (ROC) areas under the curve (AUC), it could be understood that the assessment performances of GA and HbA1c in MHD patients were lower than those in non-MHD ones, and assessment performance of HbA1c in MHD patients was better than GA (P < 0.05). In addition, the effects of Hb and EPO dose on HbA1c, or that of ALB on GA were unobvious in our study.</p><p><b>CONCLUSIONS</b>Actual glycemic control level in MHD patients with DM may be underestimated by HbA1c, and it could be avoided by GA; however, glycemic evaluating performance of HbA1c may be still better than that of GA. Therefore, HbA1c should not be replaced completely although GA can be used as a choice to monitor glycemic level.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate clinical efficacy and feasibility of double-plating fixation via anteriormiddle approach in treating type C3 distal femoral fractures.</p><p><b>METHODS</b>From August 2008 to August 2011, 12 cases with type C3 distal femoral fractures were treated, including 5 open fractures and 7 closed fractures. Among them, there were 8 males, 4 females with an average of 40 years (ranged, 25 to 55 years). There were 7 in left side, 5 in right side. Nine cases were caused by car accident, 3 cases by falling down. The duration from injury to hospital was form 20 minutes to 5 days (mean 135 min). After tibia bone traction for 5 to 8 days, the operation were performed by double-plating fixation via anteriormiddle approach, and autograft of iliac bone or allograft bone grafting were given to bone defect. Knee joint function was evaluated according to Merchanetal criteria.</p><p><b>RESULTS</b>The operation time was from 110 to 160 min, with an average of 135 min, the blood loss was from 300 ml to 500 ml,with an average of 400 ml. Post-operative wound were stage I healing. All patients were followed up from 16 to 36 months (mean 24 months). No infection, reduction loss, nonunion, deep vein thrombosis occurred. Bone healing time was for 18 to 24 weeks with an average of 21 weeks. According to the Merchanetal criteria, 4 cases got excellent results, 6 good, 1 fair and 1 poor.</p><p><b>CONCLUSION</b>Double-plating fixation via anteriormiddle approach for type C3 distal femoral fractures is an effective way, which has advantages of obvious exposure, simple manipulation, anatomical reduction, stable fixation. However,operation indications and operating instructions should be strictly followed.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Observing the dynamic change characteristics of serum liver function indexes in occupational dermatitis medicamentosa-like of trichloroethylene patients with liver damage, we can underlie for guiding therapy, prognosis and mechanism of dermatitis medicamentosa-like of trichloroethylene patients with liver damage.</p><p><b>METHODS</b>We collected serum of 10 cases of occupational dermatitis medicamentosa-like of trichloro-ethylene patients with liver damage from different time points since they were hospitalized, using automatic biochemistry analyzer to detect total protein (TP), albumin (ALB), total bilirubin (TBIL), direct bilirubin (DBIL), indirect bilirubin (IBIL), alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyl transpeptidase (GGT), alkaline phosphatase (ALP), albumin/globulin ratio etc 11 liver function biochemical indicators. We used Excel to establish database, professional drawing software gnuplot to draw dynamic variation diagram of each index.</p><p><b>RESULTS</b>The variation range of 11 liver function indexes of 10 cases was TP 43.2-74.2 g/L, ALB 24.6-44.6 g/L, A/G 0.77-2.10, TBIL 3.7-268.2 umol/L, DBIL 1.0-166.0 umol/L, IBIL 2.4 -167.5 umol/L, ALT 11-5985 U/L, AST 14-5586 U/L, GGT 15-1500 U/L, ALP 35-309 U/L, S/L 0.07-1.94, respectively. TBIL, DBIL, ALT, AST, GGT, ALP concentration significantly increased, especially ALT, AST, GGT, ALT topped 5985 U/L, AST topped 5586 U/L, GGT topped 1500 U/L. But TP, ALB and S/L significantly decreased, TP lowest to 43.2 g/L, S/L lowest to 0.07. A/G basically remained unchanged, but IBIL didn't change regularly.</p><p><b>CONCLUSION</b>The early liver damage in dermatitis medicamentosa-like of trichloroethylene patients was serious, and repeatedly attacked, so we should lead to enough attention to the clinical work and prevention. This also provided the basis for studying the mechanism of trichloroethylene poisoning.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the expression of erythropoietin (EPO) and its receptor (EPOR) in the brain of newborn rats suffering fetal distress.</p><p><b>METHODS</b>A model of fetal distress was prepared by ligating bilateral uterine arteries of the rats with full-term pregnancy for 10 minutes before cesarean sections. The expression levels of EPO and EPOR in the brain of newborn rats were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blot at 0, 2, 6, 12, 24, 48, 72 hrs and 7 days after birth. Serum EPO levels were measured using ELISA simultaneously. The newborn rats born by cesarean sections which were not subjected to uterine artery ligation were used as the control group.</p><p><b>RESULTS</b>The expression of EPO protein and mRNA in brain tissues in the fetal distress group increased significantly compared with the control group 2, 6 and 12 hrs after birth (P<0.05). The expression of EPOR protein and mRNA in brain tissues in the fetal distress group increased significantly compared with the control group 2, 6, 12, 24 and 48 hrs, and 3 days after birth (P<0.05). Serum EPO levels in the fetal distress group were significantly higher than in the control group 2 hrs after birth.</p><p><b>CONCLUSIONS</b>The EPO and EPOR levels in the brain increase quickly after birth in newborn rats suffering from fetal distress. The EPOR is high expressed for a longer time than EPO. This can provide a basis for the treatment of neonatal brain damage induced by fetal distress by exogenous EPO.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the chemosensitivity of lung adenocarcinoma cell line A549 cells to liposome-encapsulated paclitaxel after treatment by nm23-H1-small interference RNA (nm23-H1-siRNA) in vitro.</p><p><b>METHODS</b>The A549 cells were divided into two groups: non-transfected group and nm23-H1-siRNA-transfected group. Western blot analysis was used to detect the expression of nm23-H1. MTT and flow cytometry were used to determine the cell mortality rate, apoptosis rate and cell cycle after liposome-encapsulated paclitaxel treatment in both groups.</p><p><b>RESULTS</b>The expression of nm23-H1 in A549 cells was significantly decreased after transfection with nm23-H1-siRNA. After treatment for 48 hours with liposome-encapsulated paclitaxel, the cell mortality rate was increased with the increasing concentration of liposome-encapsulated paclitaxel in both groups, but increased higher in the nm23-H1-siRNA-transfected group. When the concentration of liposome-encapsulated paclitaxel was above 5 µg/ml, the cell mortality rate was significantly higher than that in the non-transfected group (P < 0.05). The proportion of apoptotic cells also increased in the nm23-H1-siRNA-transfected group, compared with that of the non-transfected group (t = 3.812, P < 0.05), while the proportion of cells at S and G(2)/M phase decreased after transfection with nm23-H1-siRNA (S phase:t = 8.356, P < 0.05; G(2)/M phase:t = 7.256, P < 0.05).</p><p><b>CONCLUSIONS</b>Nm23-H1 is related with the chemoresistance to liposome-encapsulated paclitaxel in lung adenocarcinoma cell line A549 cells. Inhibition of the expression of nm23-H1 by nm23-H1-siRNA can improve the in vitro chemosensitivity of A549 cells to liposome-encapsulated paclitaxel.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the expression of X-linked inhibitor of apoptosis protein (XIAP) in Tca8113 cell, and to investigate its relationship to the chemoresistance. METHODS; The Tca8113 cell line was cultured by IMDM and the concentration of Pingyangmycin (PYM) added to Tca8113 cell line was increased gradually and continually, which was to induce the PYM-resistance in Tca8113 cell line. The sensitivity of Tca8113 cell to PYM and expression of XIAP were measured with methyl thiazolyl tetrazolium (MTT) chromatometry and reverse transcription-polymerase chain raction (RT-PCR). The XIAP level in the cells and its chemoresistance to PYM were analyzed by linear regression.</p><p><b>RESULTS</b>The IC50 of Tca8113-1-10 group and Tca8113-10-10 group were(12.758 +/- 0.030), (18.986 +/- 0.150) microg x mL(-1) respectively. The IC50 of Tca8113-1-20 group and Tca8113-10-20 group increased to (26.302 +/- 0.072), (35.294 +/- 0.115) microg x mL(-1) respectively. There was a relation between XIAP and the drug-resistance in Tca8113 cell.</p><p><b>CONCLUSION</b>XIAP may play an important role in the chemoresistance which might serve as a new therapeutic target for oral squamous cell carcinoma.</p>

ABSTRACT

Objective To retrospectively analyze 45 cases of oral and maxillofacial malignant tumors and to assess the efficacy and feasibility of oral and maxillofacial malignant tumors implanted with 125I radioactive seeds under guidance of ultrasound.Methods A total of 47 focuses in these 45 patients were determined with the size of these tumors by imaging study,the section was planed by ultrasound,the number and distribution of radioactive seeds were determined with the help of the particle treatment planning system,and were percutaneously implanted particles under guidance of ultrasound.The number and the distribution of particles were assessed by CT.Efficacy endpoints were reexamined and evaluated regularly by ultrasonic and CT according to the standards of WHO.Results The total percentage of efficacy was 70.2% (including complete remission,partial remission).The treatment effect of metastatic carcinoma of lymph node is superior to the parotid tumor.There was no serious complication during the period of implanting and 2 patients with oral ulcers were found after operation.Conclusion The oral and maxillofacial malignant tumor treated implanted with 125I radioactive seeds under guidance of ultrasound is very effective and safe,which is deserved to popularize.The ultrasound is the first choice as a guided method for oral and maxillofacial malignant tumors.

ABSTRACT

<p><b>BACKGROUND</b>Hypertensive intracerebral hemorrhage (HICH) is a severe disease with high morbidity and mortality. Timely removal of the hematoma through surgical procedures may effectively reduce secondary injuries. However, there has long been a debate over the proper timing of such surgery. In this study, we explored the optimal operation time for HICH patients by observing the pathological changes in perihematomal brain regions during different stages of onset.</p><p><b>METHODS</b>Twenty-five specimens of brain tissue, obtained from perihematomal region of HICH patients in different phases, were subjected to haematoxylin-eosin (HE) staining, terminal deoxynucleotidyl transferase-mediated deoxyuridine 5-triphosphate nick-end labeling (TUNEL) staining and Caspase-3, matrix metalloproteinases-9 (MMP-9) immunohistochemical staining. The changing roles of necrosis and apoptosis and the expression of MMP-9 and Caspase-3 positive cells were all observed using image analysis.</p><p><b>RESULTS</b>The obvious expression of TUNEL positive cells was recognized within 6 hours of ICH onset, reaching its peak between 6 hours and 24 hours in the early phase.</p><p><b>RESULTS</b>were highly consistent with Caspase-3 and MMP-9 positive cell counts. Necrosis was found 6 hours after ICH onset and aggravated after 12 hours.</p><p><b>CONCLUSIONS</b>In the early phase, apoptosis was seen as a major modality of injury in the brain tissue of the perihematomal region and was strongly correlated with the expression of MMP-9 and Caspase-3. The results of the present study suggest that an operation performed as soon as possible after ICH onset may be optimal for preserving the nervous system function.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the difference of microRNA expression between HepG2 cells and L02 cells, and to identify the target genes of microRNA-18 (miR-18).</p><p><b>METHODS</b>The differentially expressed miRNAs between HepG2 cells and L02 cells were identified by miRNA chip. Target genes of miR-18 were predicted bioinformatically. Furthermore, the expression of B-cell translocation gene 2 (BTG2), a putative target gene of miR-18, was analyzed in hepatocellular carcinoma tissues and the surrounding non-cancerous tissues by RT-PCR and northern blot.</p><p><b>RESULTS</b>miR-18 was over-expressed in HepG2 cells compared to L02 cells. Altogether 609 genes, including genes involved in cell proliferation, differentiation, apoptosis and transcriptional regulation, are identified as putative miR-18 targets. The mRNA level of BTG2 was much lower in hepatocellular carcinoma tissues than in the corresponding non-cancerous tissues.</p><p><b>CONCLUSION</b>miR-18 is over-expressed in HepG2 cells compared to L02 cells, and it may negatively regulate the expression of BTG2, a tumor suppressor gene.</p>
